Description

Reglera uttrycket av proteiner med en vektor som bär den inducerbara tetracyklinpromotorn/operatorn. IBA Lifesciences GmbH E. coli expressionsvektor utan sekretion möjliggör expression av proteiner i E.coli.

  • pASG-IBA-vektorerna är designade för uttryck av målproteiner utan affinitetsmärkning i E. coli .
  • Vektorn bär en ampicillinresistenskassett för selektion av transformerade E. coli -celler, F1- och ColE1-ursprung för ett högt plasmidkopietal och den inducerbara tetracyklinpromotorn/operatorn för det reglerade uttrycket av proteiner.
  • Vektorerna kan kombineras med vilken E. coli -stam som helst eftersom tet-promotorn fungerar oberoende av den genetiska bakgrunden för E. coli .
  • Expression av det rekombinanta proteinet induceras genom tillsats av anhydrotetracyklin, som är ett derivat av tetracyklin som inte uppvisar antibiotisk aktivitet.
Observera att kloning i pASG-IBA Acceptor Vectors obligatoriskt kräver restriktionsenzymet Esp3I eftersom ingen annan MCS för integrering av en gen av intresse är tillgänglig. Förutom direkt kloning av genen av intresse i pASG-IBA-vektorer med Esp3I, är ett annat alternativ via en så kallad Entry-Vector möjlig.
