CD40 Ligand (CD40-L), or CD154, is a membrane glycoprotein and differentiation antigen expressed on the surface of T cells. The CD40 Ligand stimulates B cell proliferation and secretion of all immunoglobulin isotypes in the presence of cytokines. CD40 Ligand has been shown to induce cytokine production and tumoricidal activity in peripheral blood monocytes. It also co-stimulates proliferation of activated T cells and this is accompanied by the production of IFN-gamma, TNF-alpha, and IL-2. Description: CD40 ligand, (CD40L, also known as CD154, TRAP or gp39) is a membrane glycoprotein expressed on activated CD4+ T-cells, NK cells, mast cells, basophils and eosinophils. The CD40-CD40L interaction stimulates B cell immune response which includes cell surface antigen expression, cell cycle activation, Ig isotype switching, Ig secretion and memory generation. The CD40-CD40L interaction also plays important roles in monocyte and dendritic cell activation, T-cell co-stimulation and cytokine production. It has been reported that the CD40-CD40L interaction is involved in the pathogenesis of amyloid pathology in Alzheimer disease. Applications Reported: This Human CD154 (CD40 Ligand) Recombinant Protein is biologically active and can induce IL-8 secretion by normal human peripheral blood cells. Applications Tested: The activity of this protein was determined by measuring IL-8 production by normal human peripheral blood cells. Source: E. coli (amino acids Gln114-Leu261; accession # NP_000065.1). Bioactivity: The ED50 of this protein, as measured by stimulation of IL-8 production by normal human peripheral blood cells, is less than or equal to 5-10ng/mL. Endotoxin: Less than 0.1ng/ug cytokine as determined by the LAL assay. Purity: >98% as determined by SDS-PAGE and HPLC. Molecular Weight: 16.3 kDa. Storage and handling: For best recovery, quick spin vial prior to opening. Use in sterile environment. Reconstitute in 5-10 mM sodium phosphate, pH 7.5 to a concentration of 0.1-1mg/mL. Do not vortex. Store lyophilized protein at less than or equal to -20°C.
