Description
Tumor Necrosis Factor-Alpha (TNF alpha) is a protein secreted by lipopolysaccharide-stimulated macrophages, and causes tumor necrosis when injected into tumor-bearing mice. TNF alpha is believed to mediate pathogenic shock and tissue injury associated with endotoxemia. TNF alpha exists as a multimer of two, three, or five noncovalently linked units, but shows a single ∽17kDa band following SDS-PAGE under non-reducing conditions. Description: Human TNF-alpha is a 17kDa factor produced by macrophages, monocytes, neutrophils, CD4^+ T cells and NK cells. A 26kDa form of TNF-alpha is expressed as a membrane bound molecule. TNF-alpha is cytolytic and plays an important role in immune regulation. Dimers and trimers of TNF-alpha have been observed. Applications Reported: Recombinant human TNF alpha is biologically active and can induce cytolysis of L929 mouse cell line in culture. Applications Tested: The ED50 of this protein, as measured by L929 cytotoxicity assay, is less than or equal to 80 pg/mL. This corresponds to a specific activity of greater than or equal to 1.3 x 10e7 Units/mg. Source: E. coli expressed amino acids Val 77-Leu 233 of human TNF alpha accession # HUMTNFAB, N-terminal His-tag. Bioactivity: The ED50 of this protein, as measured by L929 cytotoxicity assay, is less than or equal to 80 pg/mL. This corresponds to a specific activity of greater than or equal to 1.3 x 10e7 Units/mg. Endotoxin: Less than 0.01ng/ug cytokine as determined by the LAL assay. Purity: >95% as determined by SDS-PAGE. Molecular Weight: 19 kDa. Storage and handling: For best recovery, quick-spin vial prior to opening. Use in a sterile environment. Purity: Greater than 90%, as determined by SDS-PAGE. Aggregation: Less than 10%, as determined by HPLC. Filtration: 0.2μm post-manufacturing filtered.Specifications
