Alla primära antikroppar
Primära antikroppar, immunglobuliner som binder till specifika proteiner eller andra biomolekyler, används i många forskningsapplikationer och protokoll för att upptäcka mål av intresse. De utvecklas med hjälp av olika djurvärdar, inklusive mus, råtta, kanin, get, får och många andra.
Monoklonala och
polyklonala antikroppar En typ av primära antikroppar, monoklonala antikroppar, ger hög reproducerbarhet och låg korsreaktivitet och bakgrundsljud. En annan typ, polyklonala antikroppar, kostar ofta mindre och ger större affinitet och snabbare bindning. Båda produceras med plasma B-celler, men den förra använder samma klon och den senare använder olika kloner. Monoklonala antikroppar kräver hybridomcellinjer, och polyklonala antikroppar gör det inte.
Det finns också rekombinanta monoklonala antikroppar med liknande fördelar, som hög affinitet, skalbarhet och specificitet. De tillverkas med hjälp av in vitro kloning av plasma B-celler och expressionsvärdar.
Konjugerade
primära antikroppar Antikroppar kan märkas med olika fluoroforer eller detektionsmedel eller användas utan märkningar. Märkta primära antikroppar, även kända som konjugerade primära antikroppar, hjälper forskare att förenkla och effektivisera sina applikationer. De är kopplade med vanliga enzymer och färgämnen som Alexa Fluor och används ofta i protein- och cellanalys.
Tillämpningar Antikroppar för biovetenskapliga tillämpningar används i flödescytometri, western blotting, ELISA, immunhistokemi och immuncytokemi. Sekundära antikroppar kan tillsättas för att stödja detektion och rening av vissa antigener. De binder till den primära antikroppen, som binder till antigenet av intresse. Att hitta rätt kombination av antikroppar kan resultera i större antigenspecificitet och en stark, detekterbar signal.

| Testspecificitet | Recognizes a disaccharide epitope, Gal1-3GalNAc, of Thomsen-Friedenreich (TF) antigen. It is specific for both anomeric forms of the disaccharide (TF and TF, including related structures on the glycolipid) and shows no cross-reactivity with sialylated glycophorin. The Thomsen-Friedenreich antigen acts as an oncofetal antigen, with low expression in normal adult tissues but increasing to fetal levels of expression in hyperplasia or malignancy. It is considered as a pan-carcinoma marker. This MAb is capable to agglutinate desialylated red blood cells. During metastasis, the ability of malignant cells to form multicellular aggregates via homotypic or heterotypic aggregation and their adhesion to the endothelium are critical. The tumor-associated carbohydrate Thomsen-Friedenreich antigen (Gal-GalNAc) is involved in tumor cell adhesion and tissue invasion. It also causes an immune response, and overexpression of the antigen causes cancer cells to be more sensitive to natural killer cell lysis. The Thomsen-Friedenreich antigen is suppressed in normal healthy cells and represents one of the few chemically well-defined antigens associated with tumor malignancy. The presence of the Thomsen-Friedenreich antigen on the surface of cancer cells may result from a divergence from the normal pathway for O-linked glycosylation in these cells, most likely caused by inappropriate localization of the enzymes involved in synthesis of the disaccharide. |
