Phosphatidylinositol 3-kinase (PI3K) is composed of a catalytic subunit (p110δ) and a regulatory subunit (p85α in this case), which are encoded by the PIK3CD and PIK3R1 genes. PI3K phosphorylates phosphoinositides on the 3-hydroxyl group of the inositol ring. It plays a role in oncogenic transformation and is consistently expressed at a high level in AML blast cells. PI3K can play a selective role in the amplification of phosphatidylinositol (3,4,5) trisphosphate (PIP3) levels that leads to neutrophil polarization, and regulates directional migration, but not random locomotion. The PI3K signaling pathway is involved in immune responses relevant to the pathogenesis of rheumatoid arthritis and other inflammatory diseases.

    PIK3CD/PIK3R1 (p110 delta/p85 alpha) was diluted in assay buffer and the activity determined in a 10μL Adapta™ assay (PV5099) with 10μM ATP and 50μM PIP2:PS lipid kinase substrate (PV5100) at room temperature for 1 hour. The reaction was stopped with a 5μL addition of EDTA and detection reagents. The final concentrations are 10mM EDTA, 2nM Adapta™ Eu-anti-ADP antibody and 4nM Alexa Fluor™ 647 ADP Tracer. The plate was read after a 45 min. room temperature incubation
